The increased work load associated with the development and implementation of fisheries managed under ASMFC Fishery Management Plans (FMP) has resulted in increased demands upon staff time. These FMP's require additional data collection, permits and exchange of information. Ideally, additional permanent staff would have been hired to handle the increased workload, however, this could not be done because of hiring restrictions for permanent staff. The department has chosen to employee temporary staff to assist with ongoing projects and to issue permits required under the various fishery management plans. This report will summarize those activities initiated under during the original grant period and for work planned during two extension periods for this grant. Our primary focus was on wood-associated animal species and their microbiomes. We collected, identified, georeferenced, vouchered, and subsampled these animals, cultivated their associated bacterial symbionts, and sequenced the symbiont genomes and host metagenomes. We performed phylogenetic and bioinformatic analyses to identify these organisms and their associated antimicrobial compounds and hydrolytic enzymes capable of degrading the components of wood (lignocellulose). We then performed biochemical and growth inhibition assays to confirm the activities of these compounds. We discovered a new antibiotic with activity against Acinetobacter, an emerging multidrug-resistant pathogen of growing public health concern. We described and formally named one new bivalve genus and species and four new microbial species and submitted voucher specimens and isolates to appropriate public collections. We discovered and isolated at least 25 new bacterial species and hundreds of new bacterial strains and deposited them to the publicly accessible Ocean Genome Legacy biorepository. We showed that these bacteria, now available for further characterization, have great potential for drug and enzyme discovery. We determined the mechanism by which wood-boring bivalves (shipworms) produce and export cellulolytic enzymes and possibly antimicrobial compounds made by their symbiotic bacteria. Finally, we generated 3D maps of the site using photogrammetry and sidescan sonar. Our research demonstrated that marine wood deposits, like the Alabama undersea forest, are rich sources for discovering new animal and bacterial species and associated compounds of pharmaceutical and industrial value. Previous survey efforts focused on one sea basin at a time, with surveys conducted in the Bering Sea in 2012 and 2013, the Chukchi Sea in 2016, and the Beaufort Sea in 2021. In 2025, the Alaska Fisheries Science Center’s Polar Ecosystems Program conducted flights over the Bering, Chukchi, and Beaufort seas to survey available sea ice habitat throughout these species’ geographic range in U.S. waters in a single season. Here, we investigated which functional species traits (scale type, schooling behavior, and position-in-water-column) are detected by eDNA metabarcoding and beach seines in nearshore eelgrass, mixed eelgrass, and understory kelp habitats. Using data from 35 sites across southeast Alaska, we applied occupancy modeling to estimate detection of species traits by each method. Detection probability with eDNA was 27 times greater for the species with deciduous scales (Clupea pallasii) compared to species with non-deciduous scales, and lower for species with plates (rather than scales). Conversely, species with plates showed greater odds of detection with beach seines. Given the novelty of eDNA sampling, quantifying interactions between detection and functional traits will be important to accurately characterize species distributions across marine habitats. AB - We revise the diagnosis of the flathead genus Elates Jordan & Seale, 1907 and describe a new species based on specimens purchased from a fish market in the southern Philippines. The new species differs from the only previously valid species in the genus, Elates ransonnettii (Steindachner, 1876), in having a large eye, a broad lateral vomerine tooth patch, a medial vomerine tooth patch, two pores on each lateral-line scale, a greater number of scale rows above the lateral line, an absence of a filamentous ray extending from the upper lobe of the caudal fin, a transparent dorsal-fin membrane with no dark pigmentation, and a wholly dark peritoneum. Analysis of cytochrome c oxidase subunit I (COI) data supports both genetic differentiation between E. ransonnettii and the new species as well as monophyly of the genus Elates. The new species is known only from specimens purchased in the General Santos Public Market in General Santos City caught in Sarangani Bay, in contrast to E. ransonnettii, which is broadly distributed in the western Pacific Ocean to the Malacca Strait and Andaman Sea.
