Description:This report focuses on the population genetics and systematics of the hard shell clam "Mercenaria", an important aquaculture species, as determined by restriction enzyme digestion of mitochondrial DNA (mtDNA). Fifteen clam populations were sampled along the east and Gulf coasts of the United States and assayed for mitochondrial genotype to determine the extent of geographic differentiation and thus evaluate the suitability of current hard shell clam breeding practices. A phylogenetic analysis was conducted on the clam taxa "M. mercenaria, M. campechiensis" and "M. mercenaria texana" based on the information derived from mtDNA variation. The implication of this analysis is that considerable genetic divergence has occurred between these taxa which validates the use of `trait-​specific' breeding approaches involving hybridization of the closely related species "M. mercenaria" and "M. campechiensis". This analysis also indicated that the "texana" group may be of multiple maternal origin and in all probability deserves species distinction separate from "M. mercenaria".
